CARBOHYDRATE UTILISATION BROTH

A basal medium for the performance of carbohydrate fermentation studies. Conforms with Australian Standard Methods.

Description

Amyl Carbohydrate Utilisation Broth, AM 723, is formulated in accordance with AS 1766.5-19941 and, when supplemented with the appropriate carbohydrate, can be used as part of a systematic procedure for the identification of a wide range of bacteria. The carbohydrate free basal medium is capable of supporting the growth of fastidious and less fastidious organisms which in the presence of the required carbohydrate, organisms capable of utilising that carbohydrate will produce acid or acid and gas, thereby altering the pH of the medium and causing a colour change of the medium from purple at pH 6.8 to yellow at pH 5.2.

Although a number of carbohydrates are capable of withstanding normal heat sterilisation temperatures, many may have their structure altered by these procedures and, it is therefore preferable that a filter sterilised solution of the carbohydrate is added to the sterilised basal medium and aseptically distributed as outlined in the preparation procedure.

If very fastidious organisms are to be inoculated into the medium, a small amount of sterile rabbit or bovine serum, usually to a level of 5%, can be added prior to dispensing into inoculation tubes2. Cultivation of anaerobic organisms can be enhanced by the addition of a small amount of agar, e.g. 0.1-0.2%, prior to sterilising the basal medium, creating a ‘sloppy’ agar that inhibits oxygen penetration.
