Description

Sperber and Deibel developed and described an accelerated enrichment serology procedure for the detection of Salmonella in dried foods and feeds. The method postulated an 18 hour pre-enrichment, a 24 hour selective enrichment, a 6-8 hour elective enrichment and a 2 hour serology test. In the elective enrichment step, mannose was substituted for dextrose; an ingredient in APT Broth cited in the traditional procedure for Salmonella clearance found in the Bacteriological Analytical Manual. The incorporation of mannose in place of dextrose overcame the problem of non-specific fimbrial agglutination in the serology procedure. M Broth – the revised formulation contained all the necessary nutrients to produce rapid growth of Salmonella and development of flagella, where citrate supplied the energy source previously provided by dextrose. Fantasia, Sperber and Deibel conducted a comparison study utilising the traditional method for Salmonella detection (BAM) versus the accelerated method (ES) and found that a more speedy product clearance was obtained whilst accuracy and sensitivity were maintained.
