Description
Mannitol Selenite Cystine Broth is modified from the formula of Leifson, with added cystine and is recommended for use in the examination of food for Salmonella by Standards Association of Australia (AS 1766.2.5, 1983), in conjunction with Tetrathionate Broth.
Leifson noted that Selenite medium performed most efficiently under reduced oxygen tension. The addition of cystine gives a very good recovery of Salmonella. This may be due to the reducing properties of cystine which lowers toxicity of selenite and/or the extra organic sulphur in cystine may have a restrained effect on the critical sulphur components of the bacteria, which in turn reduces the selective effect of the Selenite.
Mannitol Selenite Cystine Broth is a suitable enrichment broth in the isolation of Salmonella from faeces, foodstuffs – particularly egg products and other samples.
