PERFRINGENS SUPPLEMENTS, OPSP1 and OPSP2

Antimicrobial supplements which, when combined together and incorporated into Amyl Perfringens OPSP AgarBase, AM 152, produce a selective medium for the identification and enumeration of Clostridium perfringens from foods.

Description

Amyl OPSP Perfringens Agar is based upon the formulation of Handford1, who, recognizing the problems associated with the identification and enumeration of Clostridium perfringens in foodstuffs, developed a buffered, egg free medium more suitable for the purpose.

Selectivity of the medium for Cl.perfringens is achieved by the presence of 100μg/ml of Sulphadiazine, 0.5μg/ml of Oleandomycin and 10 IU per ml of Polymixin B, the combination of which inhibit the growth of other sulphite reducing organisms, particularly members of the Proteus and Citrobacter genera, as well as the majority of gram negative organisms together with gram positive organisms such as Staphylococcus sp. and Bacillus sp. Other sulphite reducing anaerobes such as Cl.bifermentans, the presence of which could interfere with the detection and enumeration of Cl.perfringens, are also inhibited. Not inhibited are enterococci which appear as small, white colonies, easily distinguishable from the black colonies formed by Cl.perfringens.
