SABOURAUD DEXTROSE AGAR

For the growth maintenance and identification of fungi.

Description

The low pH and high glucose content of Amyl Sabouraud Dextrose Agar, AM171,makes it selective for fungi, yeasts, aciduric and acidophilic bacteria. The inclusion of Amyl Mycological Peptone RM307), encourages luxuriant growth of dermatophytes, saprophytic moulds and yeasts, allowing rapid development while stimulating the formation of typical morphology and pigmentation. Amyl Bacteriological Agar (RM250), incorporated to a concentration of 14g/L makes Amyl Sabouraud Agar especially suitable for use either in petri dishes, or in bottles or tubes as agar slopes.

The addition of antibiotics renders the medium more selective for yeasts or pathogenic fungi, retarding the growth of saprophytic fungi or bacteria associated with inoculum from heavily contaminated material. Antibiotics may be used singly or in combination.

With the exception of chloramphenicol, gentamicin and cycloheximide, antibiotics should be added as solutions to the sterilised liquid medium, cooled to 45°C prior to dispensing. Chloramphenicol, gentamicin and cycloheximide may be added before autoclaving.

For the differentiation of Candida albicans aseptically add 10 ml of a 1% filtered sterile solution of triphenyltetrazolium chloride to 1 litre of sterile Sabouraud Medium cooled to 45°-50°C.
