Description
Y.M Broth and Y.M.Agar are used for the detection, isolation and cultivation of yeasts, moulds and aciduric organisms.
The formula follows the original papers by Wickerham.1,2
Various techniques have been developed for the use of the media. Originally, the pH of the media was reduced to pH 3-4 to make the formula selective. It must be remembered that if this is done with the agar, it must be autoclaved first at its higher pH, and then cooled to approximately 50°C and prior to acidifying by the addition of 10% Lactic acid, SP 431. THIS MUST NOT BE DONE BEFORE AUTOCLAVING
If the form being used is a broth, then this can be acidified before autoclaving.
